Workflow: Automated Scratch Assay with the Scratch Maker & zenCELL owl

Performing a reproducible cell migration or wound healing assay has never been easier.
Follow these simple steps to achieve consistent, high-quality results with the Scratch Maker and innoME pre-coated culture plates.

1. Preparation

Unpack the innoME culture plates, which come pre-coated for adherent cells and sealed in dark, sterile foil bags to ensure maximum cleanliness and coating stability.
The plates are ready to use – no additional coating or surface treatment required.

2. Cell Cultivation

Seed your cells according to your standard protocol and cultivate them under optimal conditions for X days, depending on the cell line and assay requirements.
The uniform surface of the innoME plates ensures even cell adhesion and growth, providing an ideal monolayer for scratch assays.

3. Scratch Creation

Once the desired confluency is reached, place the scratch mask (included in the starter bundle) on top of the culture plate and insert it into the Scratch Maker.

Start the light-based scratch process and wait approximately one minute.
The system creates uniform, contact-free scratches using controlled illumination – ensuring 100% reproducibility and no mechanical cell damage.

4. Incubation & Live Monitoring

After the scratch process is complete, remove the plate and place it back into the incubator.
From here, the zenCELL owl live-cell imager can continuously monitor cell migration in real time – directly inside the incubator.

With its 24 integrated cameras, zenCELL owl captures time-lapse images and AI-based cell analysis automatically, giving you complete documentation of wound closure and cell movement.

5. Data Evaluation

Using the zenCELL owl software, evaluate cell migration speed, closure rate, and reproducibility across all wells.
Export high-quality image series and quantitative data for publication, QA, or further analysis.
